Bu \u00e7al\u0131\u015fmada, yeni bir besiyerinin etkinli\u011finin NNN besiyeri ve molek\u00fcler y\u00f6ntemle kar\u015f\u0131la\u015ft\u0131r\u0131lmas\u0131 ama\u00e7lanm\u0131\u015ft\u0131r.<\/p>\n<p><strong>Y\u00f6ntemler<\/strong>: Manisa Celal Bayar \u00dcniversitesi T\u0131p Fak\u00fcltesi T\u0131bbi Parazitoloji Anabilim Dal\u0131\u2019na Kilis, Osmaniye, Bitlis, Gaziantep ve Manisa illerimizdeki sa\u011fl\u0131k merkezlerinden tan\u0131 veya do\u011frulama amac\u0131yla kutan\u00f6z lay\u015fmanyaz (KL) \u015f\u00fcpheli 22 ve viseral lay\u015fmanyaz (VL) \u015f\u00fcpheli 4 hasta \u00f6rne\u011fi g\u00f6nderilmi\u015ftir. Bu illerdeki sa\u011fl\u0131k merkezlerine \u00f6nceden haz\u0131rlanan besiyerleri g\u00f6nderilmi\u015f ve al\u0131nan klinik \u00f6rneklerden bu besiyerlerine ekim yap\u0131lmas\u0131 istenmi\u015ftir. KL i\u00e7in al\u0131nan i\u011fne aspirasyon s\u0131v\u0131s\u0131 ve VL i\u00e7in kemik ili\u011fi \u00f6rnekleri, NNN besiyerine ve yeni geli\u015ftirilen karaci\u011fer ekstreli besiyerlerine e\u015fzamanl\u0131 olarak ekilmi\u015f, yayma preparatlar\u0131yla birlikte kurumumuza g\u00f6nderilmi\u015ftir. Yayma preparatlar\u0131 Giemsa y\u00f6ntemiyle boyanarak incelenmi\u015ftir. Ayr\u0131ca bu materyallerde ger\u00e7ek zamanl\u0131 polimeraz zincir reaksiyonu y\u00f6ntemiyle parazit DNA\u2019s\u0131 aranm\u0131\u015ft\u0131r.<\/p>\n<p><strong>Bulgular<\/strong>: 26 \u00f6rne\u011fin 14\u2019\u00fcnde Leishmania amastigotlar\u0131 g\u00f6r\u00fclm\u00fc\u015ft\u00fcr. \u00d6rneklerden NNN besiyerine ekilenlerin 8\u2019inde, yeni besiyerine ekilenlerin ise 24\u2019\u00fcnde promastigot \u00fcremesi g\u00f6r\u00fclm\u00fc\u015ft\u00fcr. KL \u015f\u00fcpheli 2 hastada her iki besiyerinde de parazit \u00fcretilememi\u015ftir. Genotiplendirme yap\u0131ld\u0131\u011f\u0131nda dermotropik yerle\u015fim g\u00f6steren izolatlar\u0131n L. tropica, L. major ve L. infantum\/donovani; viserotropik yerle\u015fim g\u00f6steren izolatlar\u0131n ise L. tropica ve L. infantum\/donovani oldu\u011fu saptanm\u0131\u015ft\u0131r.<\/p>\n<p><strong>Sonu\u00e7lar<\/strong>: Lay\u015fmanyaz tan\u0131s\u0131nda yayg\u0131n olarak kullan\u0131lan NNN besiyeri \u00fclkemizde baz\u0131 Leishmania su\u015flar\u0131n\u0131n tan\u0131s\u0131nda yeterli duyarl\u0131l\u0131kta de\u011fildir.
